Usage Copovidone, an analog of povidone, is used as a tablet binder, a film-former, and as part of the matrix material used in controlled-release formulations.


2. Description:

Copovidone is a linear copolymer of N‑vinylpyrrolidone (NVP) and vinyl acetate (VA), usually in a mass ratio of 60:40. It combines the excellent properties of Povidone (PVP) with low hygroscopicity, good solubility, and film-forming ability.

Properties:Since its structure contains both NVP and VA units, it exhibits the combined characteristics of PVP and PVAC. In comparison with these two polymers, copovidone retains the good water solubility, adhesion and film-forming properties of PVP, while having significantly lower hygroscopicity, wider solubility, better plasticity and stronger surface activity. It shows higher affinity for hydrophobic surfaces than PVP.


Therefore, this product is a kind of fine tablet binder, which has the characteristics of high hardness and low brittleness, and can be used in the wet conditions. It is especially suitable for high dose, poor water solubility and water sensitive drug. 


3. Application:

To improve the in vitro solubility of tanshinone ⅡA solid dispersions and reduce their hygroscopicity, copovidone was used as a carrier to prepare tanshinone ⅡA solid dispersions.

Solid dispersions prepared with copovidone as the carrier can significantly enhance the dissolution of tanshinone ⅡA, while exhibiting relatively low hygroscopicity and good stability.

This effect can be attributed to the unique polymeric structure of copovidone, a linear copolymer of N‑vinylpyrrolidone and vinyl acetate (PVP/VA). Compared with povidone (PVP), copovidone possesses stronger surface activity and superior solubilization performance, thereby more effectively promoting the dissolution of tanshinone ⅡA.

PVP K30 is highly hygroscopic, whereas the vinyl acetate moieties in PVP/VA introduce a certain degree of hydrophobicity, lowering the overall hygroscopicity of the copolymer and improving the storage stability of solid dispersions.

As a 60:40 copolymer of vinylpyrrolidone and vinyl acetate, copovidone shows good solubility and low hygroscopicity. Using copovidone as a carrier in the preparation of tanshinone ⅡA solid dispersions can reduce wetting during processing, lower product moisture content, enhance the stability of tanshinone ⅡA within the carrier system, and retard aging of solid dispersions, which is favorable for the industrial application of solid dispersion technology.

In most cases, its dosage accounts for 2% - 5% of the micro-gross preparation, typically around 3%, with the adhesive concentration generally ranging from 5% to 10%. Depending on the different main materials and auxiliary materials, solvents such as water or alcohol can be used.

For direct compression, the recommended dosage is approximately 5%. It can be used in combination with excipients such as lactose, sorbitol, mannitol, and microcrystalline cellulose. However, it is important to control the moisture content in the ingredients to maintain appropriate bonding performance.
